Effects of Actinomycin D on Ultrastructure of Rat Hepatocytes


Abstract
The experiments was carried out to clarify effects of actinomycin D on ultrastructural alterations in the hepatic cells of rats.
A single intramusclar injection, 0. 5pg of the drug per gm. of body weight, were made on the three groups of rats. They were killed separately at the first, second and fifth days after the injections. The liver specimens obtained from each group were. observed by electron-microscope. Control studies on the changes in normal cells were conducted by sacrificing four normal rats.
It was found that in the experimental groups, the rER are fragmented, distorted, and dilated at first day after injection. The ribosomes detached out from the rER membrane are destructed from the first day observations, and on the second day these findings are obviously exacerbated. Then it has disappeared on the fifth day. The Golgi complex are proliferated on the second day and reached to the maximal extend on the fifth day.
These results suggested that actinomycin D may evoke the marked degenerative changes in the organelles of the liver cells, but the changes have disappeared from the second day to the fifth day.
